Nuclear pore complexes (NPCs) play a critical role in maintaining the equilibrium between the nucleus and cytoplasm, enabling bidirectional transport across the nuclear envelope, and are essential for proper nuclear organization and gene regulation. Perturbations in the regulatory mechanisms governing NPCs and nuclear envelope homeostasis have been implicated in the pathogenesis of several neurodegenerative diseases. The ESCRT-III pathway emerges as a critical player in the surveillance and preservation of well-assembled, functional NPCs, as well as nuclear envelope sealing. Recent studies have provided insights into the involvement of nuclear ESCRT-III in the selective reduction of specific nucleoporins associated with neurodegenerative pathologies. Thus, maintaining quality control of the nuclear envelope and NPCs represents a pivotal element in the pathological cascade leading to neurodegenerative diseases. This review describes the constituents of the nuclear-cytoplasmic transport machinery, encompassing the nuclear envelope, NPC, and ESCRT proteins, and how their structural and functional alterations contribute to the development of neurodegenerative diseases.

Pinecone oil supplemented to multiparous sows from 107 days prenatal to 21 days postpartum improves reproductive performance and milk composition and affects serum parameters

Abstract

Pinecone oil (PO) of Pinus koraiensis mainly contains α-pinene, β-pinene, and limonene that may ameliorate animal well-being and growth performance. This study evaluated its effects on feed intake, milk composition and yield, serum parameters, and litter growth of sows. Twenty-seven pregnant sows (parity 2–4) were distributed to three dietary treatments. The trial started on Day 107 of gestation and ended on Day 21 of lactation. Sows were given either a basal diet or the basal diet + 200 or 400 mg/kg PO. Each treatment contained nine sows and each sow was considered an experimental unit. Results showed that the average daily gain and weaned body weight of piglets from the sows fed 400 mg/kg PO supplements were higher (p < 0.05) than the piglets from the control sows. Lactose content in colostrum samples and fat content in milk samples were higher (p < 0.05) in 400 mg/kg PO-treated sows, respectively, than those from the sows fed basal diet. Additionally, cortisol concentration and aspartate aminotransferase concentration in sow serum was lowered (p < 0.05) by 400 mg/kg PO on Day 21 of lactation. In conclusion, supplementation of 400 mg/kg PO during late gestation and lactation contributed to greater offspring growth performance, possibly by enhanced milk quality and alleviated maternal stress.

In ovo feeding of vitamins in broilers: A comprehensive meta‐analysis of hatchability and growth performance

Abstract

In ovo feeding has been introduced as a cost-effective method to improve hatchability and broiler performance. Specifically, several studies have focused on the impact of vitamins. However, due to variations in experimental conditions across all trials, drawing general conclusions appears challenging. Therefore, we conducted a meta-analysis of 17 published papers, including a maximum of 134 sample size to evaluate the potential effects of in ovo feeding of vitamins in broilers. Studies were retrieved by consulting scientific repositories such as Pubmed, Scopus, Scielo, Web of Science, and Google Scholar. A binary logistic model was used to determine the parameters influencing hatchability. To assess variations in hatchling weight and growth parameters based on the vitamin category, a mixed model analysis of variance was performed, considering the study as a random effect and the vitamin category as a fixed effect. Finally, a linear mixed model was used to develop equations that explain the evolution of growth parameters based on vitamin concentration, volume, and day of injection. The results revealed that for better hatchability, it is preferable to consider heavier eggs (p = 0.007), lower volumes (p = 0.039), and late injection (p = 0.022). Vitamin E was associated with higher hatchling weight (p = 0.037), while vitamin C exhibited the lowest overall feed conversion ratio (p = 0.042). Interactions were observed between the day of injection and vitamin concentration or volume of injection for all studied growth parameters. In summary, the findings of this study suggest that hatchability during in ovo feeding is influenced by technique-related parameters, whereas growth parameters can be modulated by the category of vitamin injected. Consequently, this study lays the groundwork for future investigations assessing the effects of in ovo feeding in broilers, as it highlights the relationship between the methodology and potential outcomes.

Abstract

Recent research indicates that early detection of breast cancer (BC) is critical in achieving favorable treatment outcomes and reducing the mortality rate associated with it. With the difficulty in obtaining a balanced dataset that is primarily sourced for the diagnosis of the disease, many researchers have relied on data augmentation techniques, thereby having varying datasets with varying quality and results. The dataset we focused on in this study is crafted from SHapley Additive exPlanations (SHAP)-augmentation and random augmentation (RA) approaches to dealing with imbalanced data. This was carried out on the Wisconsin BC dataset and the effectiveness of this approach to the diagnosis of BC was checked using six machine-learning algorithms. RA synthetically generated some parts of the dataset while SHAP helped in assessing the quality of the attributes, which were selected and used for the training of the models. The result from our analysis shows that the performance of the models used generally increased to more than 3% for most of the models using the dataset obtained by the integration of SHAP and RA. Additionally, after diagnosis, it is important to focus on providing quality care to ensure the best possible outcomes for patients. The need for proper management of the disease state is crucial so as to reduce the recurrence of the disease and other associated complications. Thus the interpretability provided by SHAP enlightens the management strategies in this study focusing on the quality of care given to the patient and how timely the care is.

The normal relationship between fat and lean mass for mature (21–30 year old) physically fit men and women

Abstract

Objective

Determine if relative body fat (%BF) remains a biological norm in physically active, non-obese American men and women and determine reference values for other components of body composition.

Methods

Participants (n = 174 men, 70 women) were physically fit U.S. Marine 2nd Lieutenants, in their third decade of physical maturity (age 21–30). Body composition was assessed by dual-energy x-ray absorptiometry (DXA) and bioelectrical impedance analysis (BIA); and body images were obtained by 3D body scans.

Results

For men and women, respectively, %BF averaged 16.2 ± 4.1 (median 15.3), 24.3 ± 4.5 (median 23.8); fat-free mass (FFM): 67.7 ± 7.2, 49.4 ± 5.3 kg; FFM index: 21.5 ± 1.8, 18.3 ± 1.6 kg/m2; and body mass index (BMI): 25.5 ± 1.9, 24.1 ± 2.2 kg/m2. Bone mineral content (BMC) was 5% of FFM; total body water (TBW) was 70%–72% of FFM. Physique remained similar between median and higher percentiles of %BF. Only small changes in key measures were noted across the six-month training program.

Conclusions

Mean %BF of healthy active men and women in 2021 remains very similar to the 15% and 25% posited in 1980, suggesting that relative body fat has a normal fat-lean relationship in physically mature humans. These data may bring new attention to sex-appropriate %BF.

Supplemental impact of silymarin in growing pig diet on the growth performance, total tract digestibility, faecal microflora, faecal noxious gas emission and absorption rate in blood

Abstract

The research was done to examine the impact of dietary silymarin on growth performance, total tract digestibility, faecal microbial, faecal gas emission and absorption rate in blood of growing pigs. Experiment 1: a total of 140 growing pigs (24.47 ± 2.49 kg) were used in a 6-week trial. There were four dietary treatment groups (seven replicate pens/treatment, five pigs/pen) and treatment diets composed of corn, soybean meal (SBM), distillers dried grains with solubles (DDGS), and rapeseed meal-based basal diets with 0%, 0.025%, 0.050% and 0.10% of micelle silymarin respectively. Experiment 2: A total of 18 pigs were divided into six treatment groups. Treatment diets: TRT1, TRT2 and TRT3 were basal diets with 30, 150 and 300 g powdered silymarin respectively; and TRT4, TRT5 and TRT6 were basal diets with 30, 150 and 300 g micelle-type silymarin respectively. Average daily gain (ADG) tended to increase (p < 0.10) at Week 3 and overall experiment after silymarin addition. Overall ADG and average daily feed intake are also intended to improve (p < 0.10) linearly in this study. During Week 6, growing pigs fed silymarin showed linearly increased (p < 0.05) apparent total tract digestibility (ATTD) of dry matter, nitrogen and energy. Dietary silymarin supplementation increased (p < 0.10) linearly the faecal Lactobacillus count at Week 3 while Escherichia coli count was linearly decreased at both the 3rd week (p < 0.05) and 6th week (p < 0.10). Silymarin supplementation showed no effect on faecal gas emissions. A higher (p < 0.05) absorption rate in the blood was found in micelle-type silymarin compared to powdered silymarin after the 1st, 2nd, 4th, 8th, 12th and 24th h of feeding. Results suggest that silymarin in a corn-SBM-DDGS-rapeseed meal-based diet may help to improve ADG, FI, ATTD and faecal microflora in growing pigs. And absorption rate in the blood of pig is higher in micelle-type silymarin.

Local neutrophil and eosinophil extracellular traps formation in pyodermatitis pyostomatitis vegetans

Abstract

Pyodermatitis pyostomatitis vegetans is a rare inflammatory condition, affecting the skin and/or mucous membrane. Some cases include both skin and mucous involvement, whereas others develop either skin or mucous lesions only. The typically affected areas are the scalp, face, trunk and extremities, including the flexural areas and umbilicus. Clinical features show erosive granulomatous plaques, keratotic plaques with overlying crusts and pustular lesions. Among mucous lesions, oral mucosa is most frequently involved, and gingival erythema, shallow erosions, cobblestone-like papules on the buccal mucosa or upper hard palate of the oral cavity are also observed. Some of the lesions assume a ‘snail track’ appearance. Although there are several similarities between pyodermatitis pyostomatitis vegetans and other diseases, that is pyoderma gangrenosum, pemphigus vegetans and pemphigoid vegetans, the histopathological features of pyodermatitis pyostomatitis vegetans are unique in that epidermal hyperplasia, focal acantholysis and dense inflammatory infiltrates with intraepidermal and subepidermal eosinophilic microabscesses are observed. Direct immunofluorescence findings are principally negative. Activated neutrophils are supposed to play an important role in the pathogenesis of pyodermatitis pyostomatitis vegetans. The expression of IL-36 and neutrophil extracellular traps (NETs) was observed in the lesional skin, and additionally, eosinophil extracellular traps (EETs) was detected in pyodermatitis pyostomatitis vegetans. A possible pathogenic role of NETs and EETs in the innate immunity and autoinflammatory aspects of pyodermatitis pyostomatitis vegetans was discussed.
